Laboratory of Confocal and Fluorescence Microscopy

Fluorescence bar

Laboratory Equipment

Laboratory is equipped with high-end inverted confocal microscope Leica TCS SP2 with AOBS (Acoustio-Optical Beam Splitter) system, which ensure high sensitivity and possibility to combine up to four fluroscence markers. Microscop allow structure analysis of both fixed as well as living samples. There is a chamber with CO2 atmosphere for live cell imaging available. It is possible to do multicolor analysis as well as recording of physiologial processes, 3D reconstructions, FRAP (Fluorescence Recovery After Photobleaching) and FRET (Fluorescence Resonance Energy Transfer) analysis an tile scans. There is diode 405 nm laser installed for excitation in the range near to UV.

The most up-to date confocal microscope Zeiss LSM 880 is equipped with set of lasers for one-photon excitation and with tunable  Ti:Sa pulsed laser for two-photon excitation. Hi sensitive 32-channel GaAsp spectral detector gets unrivalled clear images. It is possible to do one channel FLIM (Fluorescence Life-time IMaging), FCS (Fluorescence Correlation Spectroscopy) and RICS (Raster Image Correlation Microscopy) measurents which allow us uncover molecular relationships and functions. Microscope works with high speed and high resolution. Live cell imaging heated chamber with CO2 atmosphere is available as well.

There is also a great wide-field system Olympus Scan^R available. It is equipped with stabilized light source, high sensitive Hamamatsu camera, motorized stage, filter and objective turrets and shutters. It is posseble to do high quality imaging as well as high throughput imaging and mosaic imaging. High quality stage chamber with regulation of temperature (-5 to 65 °C) and gas level (CO2, O2) can maintain also multiwell dishes and allow high throughput imaging under different enviroment conditions.

Microscope Zeiss Elyra PS.1 use a technologies of structured illumination (SIM) and precise localization (PALM/STORM) to overcome so called diffraction barrier, which limits us to resolution of cca 200 nm in conventional light microscopy. Nanoscopy allow us observe structures with 2-3x higher resolution with SIM and up to 5 nm with localization microscopy. Microscope can use a technology of TIRF (Total Internal Reflection Fluorescence) to increase signal to noise ratio in the space near to cover glass. A set of high quality lasers and cameras help us to make great super-resolution images.

For optimal performance all microscopes stay on antivibration tables in air-conditioned rooms. Our core facility serve people of many different fields of research and contribute significantly with results published in peer-reviewed scintific journals.
